Photodynamic therapy (PDT) represents a modern approach for the treatment of diverse cancer types. This modality encompasses three fundamental components: light, oxygen, and photosensitizer. Essential requirements for an effective photosensitizer include selectivity for cancerous tissues, a distinct and precisely defined molecular structure, an appropriate spectral profile, stability at room temperature, and solubility within biological systems. While chlorins and their derivatives exhibit favorable absorption and emission characteristics for PDT, their limited solubility in biological media remains a concern. For the purpose of optimizing the pharmacological properties of chlorin-based photosensitizers to enhance their photodynamic efficacy, derivatives of chlorin e6 were synthesized, specifically 131 and 152-[N-(2-aminoethyl)maleimide] chlorin e6 derivatives. The maleimide group of chlorins was modified by cycloadditions of azide-Gu and 3-mercaptopropionic acid. Notably, the derivative incorporating 3-mercaptopropionic acid was successfully synthesized and isolated as a pure product. However, the synthesis of the azide-Gu derivative yielded a mixture of two diastereoisomers. Structural characterization of the products was achieved through various spectroscopic techniques including 1H NMR, UV-Vis, and IR spectroscopy. Furthermore, their melting points were determined to provide additional insights into their physical properties.
