Abstract
Amino acids are the building blocks used to construct peptides. Peptides can form different secondary structures such as alpha helices and beta sheets. Through evolution, these structures became more complex and formed proteins. Enzymes are proteins that attract interest in terms of scientific research. They are widely explored for their catalytic properties. They facilitate biological reactions and accelerate the reaction rate. While much is known about mechanisms of catalysis, replicating these enzymatic systems remains a challenge. One approach to address this issue is the use of short peptide sequences instead of enzymes. The advantage is possibility of manipulating their physio-chemical properties to suit specific design requirements in reactions. Self-assembly of short peptides presents a simplified method of producing successful catalysts like enzymes. Supramolecular self-assembly approach offers opportunities to manipulate such structures. However, these “de novo designed catalysts” still lack functionality compared to bigger proteins. Many scientific reports are being made about emerging strategies and possible peptide sequences that could catalyse different reactions such as ester hydrolysis, aldol reactions, deacetalization etc. Moreover, nanotechnology has evolved immensely in the past decade. Knowledge about constructing nanomaterials allowed for creating peptide nanostructures such as micelles, amyloid-like fibres and others. Catalytic ability of these formations is being tested to facilitate reactions such as ester hydrolysis. In this work, we will focus on explaining the importance of using short peptides in catalysis instead of proteins.

Aminokiseline su gradivni elementi za peptide. Peptidi formiraju različite sekundarne strukture poput alfa-heliksa i beta-ploča. Tijekom evolucije ove su strukture postale složenije i formirale proteine. Enzimi su proteini koji privlače interes u znanstvenim istraživanjima. Široko se istražuju zbog svojih katalitičkih svojstava. Oni olakšavaju i ubrzavaju biološke reakcije. Iako se mnogo zna o mehanizmu katalize, oponašanje ovih enzimskih sustava je prilično izazovno. Zbog toga se dizajniraju kratke peptidne sekvence koje se koriste za katalizu reakcija poput hidrolize estera. Njihova prednost leži u mogućnosti manipulacije njihovih fizikalno-kemijskih svojstava kako bi odgovarali specifičnim uvjetima dizajna za reakcije. Samostalno sklapanje (eng. self-assembly) kratkih peptida predstavlja jednostavnu metodu za pripremu uspješnih katalizatora kao što su enzimi. Supramolekularni pristup samostalnom sklapanju nudi mogućnosti manipulacije takvim strukturama u velikoj mjeri. Ti takozvani „novo dizajnirani katalizatori“ još uvijek nemaju funkcionalnost kakvu imaju veći proteini. Mnogo je objavljenih znanstvenih radova o novim strategijama i mogućim peptidnim sekvencama koje djeluju kao katalizatori u reakcijama poput hidrolize estera, aldolnih reakcija, deacetilacije itd. Nadalje, nanotehnologija se izrazito razvila u posljednjem desetljeću. Znanje o konstruiranju nanomaterijala omogućilo je stvaranje peptidnih struktura poput micela, vlakana sličnim amiloidima i dr. Katalitička sposobnost ovih formulacija ispituje se kako bi se olakšale reakcije poput hidrolize estera. U ovom ćemo se radu usredotočiti na važnost korištenja kratkih peptida u katalizi umjesto proteina.
